SLPI is a member of the WAP (whey acidic protein) motif family. The WAP motif is a conserved 4 disulfide sequence active in serine proteinase inhibition, and SLPI has two WAP domains. In addition, SLPI contains transglutaminase motifs that allow binding to ECM components via charged interaction of the glutamine and lysine to HS gags.SLPI is known to directly inhibit Pseudomonas aeruginosa in-vitro, and much work has been done describing SLPI as a defensin-like molecule that helps control infection. SLPI is also increased in menstruation, where it is controlled by progesterone levels, although the function is unclear.
