TACE was initially identified by its ability to cleave the membrane-anchored, 26 kDa form of TNF- and release the soluble, 17 kDa form, which induces inflammation. Subsequently, TACE has been shown to be a major sheddase of many membrane proteins involved in a variety of processes. Examples are TGF-, p75 and p55 TNF receptors, L-selectin, growth hormone receptor, amyloid precursor protein, Notch, IL-6 and type II IL-1 receptors, fractalkine,ErbB4, neuregulin, and LDL receptor. TACE is expressed in virtually every tissue.
